Sperm DNA fragmentation is a key factor in male infertility. It accurately assesses the level of DNA fragmentation, providing clinically significant diagnostic information about the potential of a patient's sperm to achieve pregnancy and eventually produce a live birth. This additional information goes above-and-beyond the traditional semen analysis.

The Sperm DNA Decondensation (SDD™) test accurately assesses the level of DNA decondensation compared to a normal fertile male. In the sequence of events following the penetration of sperm into an egg, sperm chromatin material undergoes decondensation. This is a prerequisite for DNA synthesis and proper fertilization. In one study of men with unexplained infertility, almost 22% failed the SDD™ test.All of the sperm samples that failed the SDD™ test were then used in an IVF cycle, and none produced a pregnancy that progressed beyond the first trimester.
RadioActive DNA Fragmentation Assay
The cells' DNA is radioactively labelled by growing the cells in presence of 3H-Thymidine so that radioactive 3H-Thymidine is incorporated into the DNA. Then the labelled cells are incubated with or without the DNA fragmentation inducing agent. During this incubation the added agent (e.g. TNF-alpha induces cells to die by apoptosis and consequently the fragmentation of DNA while the DNA of untreated cells remains intact. After incubation (e.g. 48 h) the cells are harvested: during harvesting, the cells are washed out of the wells of the 96 well plates with bidets water: the cells and organelles burst and the cell's DNA is set free. The cell fragments and DNA are passed through a filter membrane (glassfiber). Only particles of smaller than 1,5 µm can pass the filter.
